Ce verset aussi a été l'objet d'interprétations diverses. Jésus parle à des auditeurs qui se scandalisent de son discours. (versets 52-58) Il voudrait dissiper leurs préjugés ; il énonce, à cet effet, trois propositions :

1° C'est l'Esprit qui vivifie, le Saint-Esprit de Dieu, dont Jésus était rempli sans mesure (Jean 3.34) et qui, par sa parole, régénère les âmes et communique la vie. (Jean 3.5,Romains 8.2 ; 1Corinthiens 2.4)

2° Les paroles que je vous ai dites il y a un instant (versets 52-58) sont Esprit et vie (grec sont Esprit et sont vie ; cette double affirmation acquiert une grande énergie par la répétition du verbe), elles portent avec elles l'Esprit divin et communiquent la vie. Mais, pour cela, il faut que l'âme soit ouverte à la lumière et à la puissance vivifiante de l'Esprit, car sans lui

3° La chair ne sert de rien. Même la chair de Christ, qu'il devait donner pour la vie du monde, (verset 51) toute sa personne et toute son œuvre, qu'il s'agit de s'approprier par la foi, (versets 53,54) ne peut vivifier que par l'Esprit, qui seul fait comprendre l'incarnation et le sacrifice de Jésus-Christ et notre union avec lui. (Jean 16.14)

A ceux qui s'arrêtent à l'extérieur et ne connaissent Christ que selon la chair, (2Corinthiens 5.16) Christ lui-même ne sert de rien.

- On sait le rôle que cette parole joua dans les controverses sur la cène et en particulier dans la discussion de Luther et de Zwingle à Marbourg.

Zwingle répétait souvent les mots : La chair ne sert de rien, et Luther répondait : Jésus ne dit pas : ma chair, mais la chair, et il entend par chair la disposition charnelle du cœur corrompu de l'homme qui le porte à prendre les paroles de Christ dans un sens grossièrement littéral. (2Corinthiens 3.6) Zwingle, dans sa conception de la cène, méconnaissait le sens profond des paroles de Jésus ; (versets 53-57) Luther, de son côté, en prenant le mot chair dans un sens tout différent de celui qu'il a dans les versets précédents, méconnaissait l'évidence du contexte.
